Description : The Leiden Institute of Chemistry wishes to purchase a Spinning Disc microscope. The primary tasks of this setup is to image live zebrafish larvae (0 to 12 days post fertilization) and to image (i.e. brightfield and fluorescence imaging) biological samples (cells, organoids, tissue (sections), organs like the brain of zebrafish) before and after expansion using published Expansion Microscopy (ExM) protocols with high speed, sensitivity and resolution. Our spinning disc microscope is intended to be used to image large numbers of zebrafish larvae at different developmental stages before and after injection of nanomedicines/drugs in the blood stream or directly into tissue/organs like the brain. The microscope should be able to image this in brightfield or using fluorescence with high speed, sensitivity and resolution.   The spinning disc microscope will also be used to image cells, organoids, tissue (sections), organs like the brain of zebrafish before and after expansion of the biological sample. In the expanded state the density of fluorescent probes is very low and therefore a high sensitivity is required. Furthermore, these samples are similar to the zebrafish large in size (multiple mm tot cm in length) requiring a fastmode of imaging. The microscope will be used by light microscopy experts from the cell observatory and by selected postdoctoral students and PhD-students after receiving initial training by permanent staff with many years of experience on light microscopy.
