Other justification : The Leiden Institute of Chemistry seeks to purchase a confocal microscope for the ERC CoG-project ‘Synthetic Chimeric Antigen Receptors: Hijacking Nitrenium Ions for Targeting, Therapy and Safety of Next Generation CAR T Cell Therapy’. The project involves the live cell imaging of T cells that are modified with synthetic antigens and their interactions and killing mechanisms of target cells. Live cell confocal imaging is regularly needed to evaluate the progress and success of the project. The project makes use of fluorophores that are synthetically prepared in-house where the excitation and emission properties are difficult to predict. In addition, we expect the research to involve the use of multiple fluorophores of which the spectral detection is difficult to separate (e.g. eGFP and Fluorescein). Therefore, it is essential to access a user-friendly imaging system allowing flexible excitation wavelength settings as well as methods to separate fluorescence based on fluorescence lifetime. The STELLARIS 8 from Leica Microsystems is the only confocal platform with an integrated next-generation White Light Laser (WLL) combined with proprietary Acousto-Optical Beam Splitter (AOBS) providing an expanded excitation range from 440 to790 nm (350 laser lines) and 8 single excitation wavelengths can be selected to image multiple targets simultaneously. The Power HyD-S detector allows sensitive photocounting from 410 nm tot 850 nm range. Such a combination enables tuning excitation and detection to perfectly match the spectral profile of the dyes in our samples and giving us the flexibility to perform multicolor experiments, also in the near IR range. The expanded excitation range will also permit the use of a wider range of dyes and chemistries to track and stimulate cell-cell interaction both at the single and multicellular level. Further, the unique TauSense lifetime-based technology combined with the WLL and HyD-S detector provides photon average arrival time allowing separation of fluorophores with spectral overlap. This feature will facilitate us with the access to increasing the complexity of our functional imaging capacity. In addition, it is important for our research that the module can be extended with a digital light sheet configuration to reduce phototoxicity. Leica Microsystems is the only company that supplies a platform that combines confocal and light sheet imaging in one system which greatly facilitates experimental handling time. Additionally, Stellaris 8 provides the option and flexibility to extend the microscope with a Stimulated emission depletion (STED) module which provides the possibility to perform super-resolution imaging on living cells to study cell interactions in greater more detail. Finally, as users have a wide range of expertise an accessible and intuitive operating interface is essential. The ImageCompass smart user interface is intuitive and automatically choses optimal wavelength settings when setting up an experiment. In conclusion, Leica Microsystems is the only supplier that offers above unique features in one state-of-the-art microscope, which is necessary for the successful completion of this research project.
