LACDR wants to analyse metabolites, lipids and small molecules quantitatively with a good dynamic range and at the same identify or confirm the identity of detected features. To expand our analytical capacity, LACDR requires an additional High-Resolution Mass Spectrometer. This system must seamlessly integrate into our existing analysis workflows, support our established analytical methods, and provide results comparable to those from our current systems. Additionally, compatibility with our in-house developed software is essential. The required MS system must offer high resolution for accurate mass measurement, a wide dynamic range, Rapid MS/MS capabilities, high sensitivity for precise quantification and conventional collision induced dissociation (CID) fragmentation as well as advanced electron activated dissociation (EAD) fragmentation combined with trapping of fragment ions using Zeno trapping to characterize lipids and metabolites. This acquisition will enable LACDR to enhance its metabolomics platform, improving both quantitative analysis and feature identification. Specifically, LACDR needs a MS system meeting the requirements below: 1. Internal mass accuracy of <0.2 and <0.5 ppm for the fragment ions m/z 813 and m/z 120 of Glu-Fibrinopeptide B, respectively 2. The instrument much contain a Zeno trap to allow a duty cycle for TOF-MS/MS higher than 90% to have sufficient sensitivity, and this must be available for full-scan MS/MS, MRMHR and IDA/DDA. 3. Good sensitivity defined by the following signal-to-noise (S/N) ratios: S/N ≥ 2500:1 using an ESI probe in full-scan TOF MS mode with a mass extraction window of 10 mDa for m/z 609.2806 for a 1 pg reserpine injection on column (ESI positive mode), and S/N ≥1000:1 using an ESI probe in full-scan TOF MS mode with a mass extraction window of 10 mDa for m/z 321.0050 for a 100 fg chloramphenicol injection on column (ESI negative mode). 4. MS must offer the following acquisition modes: TOF MS/MS, MRMHR, data independent acquisition (DIA)/SWATH scan mode using fixed and variable window acquisitions, and information dependent acquisition (IDA)/data dependent acquisition (DDA) scan modes. 5. The instrument must be capable of a scan speed of 25 Hz (25 spectra per second) for the TOF MS. For the TOF MS/MS the must be capable of a scan speed of 133 Hz (133 MS/MS spectra per second). 6. The instrument must be capable of a scan speed of 133 MS/MS scans per cycle in data independent mode (DIA) or with SWATH acquisition for both fixed window and variable window methodologies. 7. The mass range of the TOF has to be up to 40kDa, and the precursor ion selection mass range 5-2,250 Da. 8. The instrument must have a maximum scan speed of up to 25 Hz (25 spectra per second) for the TOF MS. For the TOF MS/MS the must have a maximum scan speed of up to 133 Hz (133 MS/MS spectra per second). 9. The instrument must have a dynamic range of ≥5 orders of magnitude in positive and negative mode from the limit of detection (LOD) without a signal reduction for both TOF MS and TOF MS/MS 10. The instrument must have a maximum scan speed of up to 133 MS/MS scans per cycle in data independent mode (DIA) or with SWATH acquisition for both fixed window and variable window methodologies. 11. The instrument must contain a user-tuneable electron-based fragmentation mode operating at electron energies from 0 eV to 25 eV (EAD). The instrument must support full-scan TOF MS/MS, MRMHR, DIA/SWATH acquisition and IDA/DDA scan modes with EAD. The Sciex 7600 MS is the only MS that can meet all these requirements.
