The instrument must contain a time-of-flight (TOF) mass analyzer and enable sequential window acquisition of all theoretical mass spectra (SWATH) acquisition mode. The instrument must have an internal mass accuracy of <0.2 and <0.5 ppm for the fragment ions m/z 813 and m/z 120 of GluFibrinopeptide B, respectively. 4. For targeted metabolomics and proteomics, the scan speed for TOF-MS/MS must be >100 Hz to ensure the accurate quantitation of a large number of metabolites and peptides (> 300 metabolites) per run. The duty cycle (TOF-MS/MS) must be >90% for all ions, to enhance the sensitivity. The linear dynamic range must be ≥5 orders of magnitude in both ESI positive and negative modes. For both untargeted and targeted approaches, the sensitivity must be sufficiently high to allow for the detection and accurate quantitation of metabolites and peptides present at trace concentrations in biosamples. Therefore, the sensitivity must reach the following signal-to-noise (S/N) ratios: S/N ≥ 2500:1 using an ESI probe in full-scan TOF MS mode with a mass extraction window of 10 mDa for m/z 609.2806 for a 1 pg reserpine injection on column (ESI positive mode), and S/N ≥1000:1 using an ESI probe in full-scan TOF MS mode with a mass extraction window of 10 mDa for m/z 321.0050 for a 100 fg chloramphenicol injection on column (ESI negative mode).
